Comparative Studies on Chemical and Hematological Changes of Whole Blood Stored in Home Made ACD-4, ACD-B, CPD and CPDA-1

Abstract
For the evaluation of whole blood preserved in home made Plastic blood bags
containing ACD-A, ACD-B, CPD and CPDA-1, blood was drawn from a group of five
healthy male blood donors, aged 24 to 33 years, for each series of preservative solution
as seen in table 1.
After the collection, blood was kept in blood refrigerator at 1 to 6¡É.
Samples were drawn at day 0, day 1, dart 7(1w), day 14(2w), day 21(3w), day 28
(4w), day 35(5w) and day 42(6w) in 15m1 each through donor tubing with cut and
resealing under sterile conditions.
The following analysis were performed with standard procedures on each sample.
For the hematolgical study, total hemo1obin, RBC, WBC, platelet, hematocrit, MCV,
MCH, MCHC were tested. For the chemical study, ATP anA 2,3-DPG in red cells and
hemoglobin, glucose, sodium, potassium, chloride in plasma and blood pH and
bicarbonate were tested.
The results were expressed as mean of five samples with plus-minus one standard
deviation as seen in table 2, 3, 4, 5, 6, 7, 8, 9, 10 and 11.
To ascertain the sterility of each blood bag, bacterial cultures were performed after
the last sampling maneuber.
In table 12, biochemical changes of blood stored in CPD and CPDA-1 with Japanese
made CPDA-1 (K) control were shown, which are quite satisfactory in all according to
their expiration date.
In table 13, biochemical changes of blood stored in ACD-A and ACD-B were
summerized, which are also satisfactory and suggest the need of replacement of wrong
ACD-B usage in plastic blood bag in this country with ACD-A.
The platelet storage seems better in ACD-A in table 2-8 but there was no significant
difference (p>0.05).
The ATP contents of red blood cells in CPD whole blood at 3 weeks revealed 72.5%
of day 0 value which was significantly higher than its contents in ACD-A and ACD-B
(p<0.05).
The ATP contents of red blood cells in CPDA-1 whole blood at 5 weeks, 70.7% of
day 0 value, which was satisfactory and there was no significant difference between its
contents in CPDA-1(K) revealed 73.0% of day 0 value (p>0.05).
